As milk protein and lactose (also contains "milk sugar"), then no E. coli tryptophan, but would like to lactose metabolism. Therefore, the responses are scanned to see and those who are lactose operon (bacterial enzymes) metabolism of lactose and see what the responses of the trp operon is turned off and produce (the bacterium does not need the synthesis of tryptophan).
